Studies of Fluid Exchanges between Rat Liver Slices and Simple Media
نویسندگان
چکیده
1. The exchange of fluid between slices of rat liver and solutions of monosaccharides, disaccharides, and sodium chloride has been studied in relation to the temperature of incubation. The point of apparent isotonicity (P.A.I.) of the tissue was defined as the concentration of the solution in which the slices neither gained nor lost weight after immersion for a period of 10 minutes. 2. In solutions of glucose, the P.A.I. of the slices was significantly lower at 4 degrees C. than at 20 degrees C. but similar at 20 degrees C. and 37 degrees C. Upon immersion for 15 to 60 minutes in 0.66 molar glucose the slices always swelled more at 37 degrees C. than at 20 degrees C. In solutions of sucrose change in the temperature of incubation was without effect on the hydration of the tissues. 3. In solutions of sodium chloride, the P.A.I. and the content of water, chloride, and sodium plus potassium were lower at 37 degrees C. than at 20 degrees C. 4. These findings emphasize the role of translocation of solute in providing an osmotic gradient for the movement of water between the tissue slices and the media.

Modulation of Basal Glutamatergic Transmission by Nicotinic Acetylcholine Receptors in Rat Hippocampal Slices
Objective(s) Nicotinic acetylcholine receptors (nAChRs) regulate epileptiform activity and produce a sustained pro-epileptogenic action within the hippocampal slices. In the present study, we investigated the effect of nAChRs on evoked glutamatergic synaptic transmission in area CA3 and CA1 of rat hippocampal slices to identify possible excitatory circuits through which activation of nAChRs pr...
متن کاملViability, function and morphological integrity of precision-cut liver slices during prolonged incubation: Effects of culture medium.
Precision-cut liver slices (PCLS) are an ex vivo model for metabolism and toxicity studies. However, data on the maintenance of the morphological integrity of the various cell types in the slices during prolonged incubation are lacking. Therefore, our aims were to characterize morphological and functional changes in rat PCLS during five days of incubation in a rich medium, RegeneMed®, and a sta...
متن کاملStudies on carbohydrate metabolism in rat liver slices. IX. Ionic and hormonal effects on phosphorylase and glycogen.
In 1939, Ostcrn, Herbert, and Holmes (1) reported the synthesis in vitro of glycogen in rabbit liver slices incubated in a Ringer’s solution containing 1 per cent glucose and high levels of calcium ion. This observation on rabbit liver was readily confirmed; however, no net glycogen increase could be obtained in rat liver experiments under similar conditions. Since liver slices were observed to...
متن کاملA simple method for cryopreservation of liver slices from man and other species
The cryopreservation of liver slices permits optimal use of large livers and provides for the availability of the slices at any desired time. Moreover, it allows one to study liver slices from various species in the same experiment. We studied the effect of cryopreservation on the viability and metabolic activity of liver slices from rat, monkey, dog, hamster, minipig, rabbit and human. Precisi...
متن کاملThe effect of the ionic environment on the synthesis of glycogen from glucose in rat liver slices.
The effect of the ionic environment of the incubating medium upon glycogen formation by liver slices was first reported by Ostern, Herbert, and Holmes (1). Using rabbit liver slices, they found that, aerobically, calcium stimulated glycogen formation in a modified Ringer’s medium containing 1 per cent glucose. Addition of phosphate to this medium resuhed in a decrease in glycogen synthesis. The...
